Microbiological diagnostics
(objectives)
The main educational objective of this teaching is to provide the student with the basic knowledge of microbiological diagnosis, referring to basic and innovative techniques. This enables the student to gain an overview of diagnosis by understanding in which biological matrices microorganisms should be sought and what precautions to take in order to make accurate laboratory diagnoses in microbiology The main pathogens implicated in human pathogenesis and which laboratory techniques to adopt according to the pathogen will be covered. Diagnostic algorithms will be explained so that the student will be able to understand the steps of microbiological diagnosis By the end of the course, the student will have acquired the main methods and techniques to be applied in the identification of bacteria, viruses, fungi, parasites and protozoa, with the ability to apply the most appropriate diagnostic technique

-Batteri, virus e funghi di interesse medico. -Diagnostica microbiologica: campioni patologici, raccolta, trasporto, conservazione e lavorazione dei campioni. -Infezioni batteriche, esame microscopico diretto e coltura. Infezioni virali, esame diretto e coltura. -Metodi di rilevazione di macromolecole microbiche. -Test di agglutinazione al lattice. -Elisa. Immunofluorescenza. Immunoistochimica. -Diagnosi sierologica di infezione: metodi per documentare la presenza ed il titolo di anticorpi specifici. -Diagnostica microbiologica molecolare: estrazione degli acidi nucleici, metodiche di ibridazione, amplificazione del segnale di ibridazione, rivelazione post- amplificazione. -L'automazione nel laboratorio di microbiologia clinica: l’automazione in sierologia, nell'identificazione dei batteri e antibiogramma, nell'estrazione degli acidi nucleici e nei metodi di amplificazione degli acidi nucleici. -Infezioni delle vie respiratorie: i principali quadri patologici e la diagnosi di laboratorio delle infezioni delle vie respiratorio superiori e inferiori. -Le più comuni infezioni del sistema nervoso centrale (meningiti, encefaliti) e loro diagnosi di laboratorio. -Infezioni dell'apparato genito-urinario, in relazione ai principali batteri, virus e miceti coinvolti ed alle varie tipologie di diagnosi di laboratorio. Malattie sessualmente trasmesse (MST) ed infezioni delle vie urinarie (IVU). -Infezioni dell'apparato gastro-enterico: le principali infezioni dello stomaco, dell'intestino e del fegato in relazione agli agenti causali ed alla loro diagnosi di laboratorio. -Infezioni della cute, delle ossa e delle articolazioni: metodiche di diagnosi di laboratorio. -Infezioni vascolari e cardiache, batteriemie, sepsi: principali agenti infettivi e diagnosi di laboratorio. -Infezioni opportunistiche e infezioni correlate all’assistenza (ICA): definizioni, fattori di rischio, vie di trasmissione, prevenzione e trattamento.
